1.what's the difference between "outside the room" and "outside of the room"?
2.what's the difference betwen "rooster" and "cock"?

#1: no difference.  However, there could be an example where "of" may be necessary to avoid confusion in a complex sentence, though I cannot think of any example.

#2: both "rooster" and "cock" refer to the male chicken.  However, since "cock" in English also has slang meaning ( as a more vulgar form of the word "penis" ), using this word may cause an undesirable reaction when in more immature company.
